Hi Craig,
Milk Thistle works right away.  There is no real delayed action, although the longer you take it the deeper it works.  If you are taking 400mg and are not feeling anything, then you can always up the dosage.  We start people on about 400mg and raise the dosage to as much as 1800mg/day (for a short duration) depending on the situation and the client's needs.

Before you make any major decisions, however, I would consult your physician, and/or the manufacturers of the product you are using.  400mg is of total ingredients, but does not necessarily give you the level of active ingredients.  So the number, 400mg is going to provide you with a very different result from product to product.

It is all dependent on what symptoms you are currently working on and how your body normally feels.  Some take for a three month course to do a gentle liver cleanse.  Some for the rest of their lives who are battling liver disease.

The experience of taking Milk Thistle is different for most people especially in the beginning detox phase.  But after a few weeks the general response we get is the same; a feeling of being more clear, less toxic, and more in balance.  If you want to read about detox, we posted an article at: basics of detox.  Hope this helps.  Let us know if we can answer any more questions.
